Vaginal cytology is a simple, quick diagnostic procedure that examines cells collected from a female dog’s vaginal lining under a microscope. As hormone levels change throughout the estrous cycle, the appearance of these cells shifts in predictable patterns. By evaluating the types and proportions of epithelial cells present, our veterinarians can accurately identify which stage of the cycle your dog is in. According to research published in Frontiers in Veterinary Science, vaginal cytology is a routinely used and valuable diagnostic tool in canine reproduction for estrus cycle staging and the detection of gynecological conditions.
The canine estrous cycle includes four main phases. During proestrus, rising estrogen levels cause the vaginal lining to thicken, and the cells transition from small, round parabasal cells to larger intermediate and superficial cells. At estrus, the peak fertile period, superficial cornified cells dominate the sample. The shift into diestrus is marked by the sudden return of smaller cells and the appearance of white blood cells, signaling the end of the fertile window.
The process involves gently collecting a small sample of cells from the vaginal wall using a moistened cotton swab. The sample is then rolled onto a glass slide, stained, and examined under a microscope. The entire process takes only a few minutes and causes minimal discomfort to the dog. We typically recommend serial cytology exams performed every two to three days throughout the heat cycle for the most accurate results.
Vaginal cytology helps us determine the right time to begin more advanced testing, such as progesterone assays, and to schedule natural breedings or artificial insemination at the ideal moment. We also use cytology to confirm whether a dog is actually in heat, to evaluate whether it’s too late in the cycle for breeding, and to calculate expected whelping dates. Combined with our other reproductive services, cytology gives breeders a reliable roadmap for every stage of the process.
Vaginal cytology isn’t only for breeders. It also helps us diagnose infections such as vaginitis, identify abnormal hormone activity, and evaluate reproductive health in spayed dogs showing signs of estrogen influence.
